Elder, Robert L. (Johns Hopkins University, Baltimore, Md.), and Roland F. Beers, Jr. Nonphotoreactivating repair of ultraviolet light-damaged transforming deoxyribonucleic acid by Micrococcus lysodeikticus extracts. J. Bacteriol. 90:681-686. 1965.-Extracts from Micrococcus lysodeikticus repair Haemophilus influenzae transforming deoxyribonucleic acid (DNA) damaged by ultraviolet light radiation. The repair is demonstrable over a wide dose range, with a constant dose reduction factor for a given concentration of DNA. The active component in the crude extract may be separated into a heat-stable dialyzable and a heat-labile nondialyzable component. The dialyzable fraction contains at least one component which appears to limit the maximal level of repair. Mg(2+) ions are required for the repair process.

متن کاملDeoxyribonucleic acid repair in vitro by extracts of Escherichia coli.
Deoxyribonucleic acid (DNA) from bacteriophage T7 has been used to monitor the capacity of gently lysed extracts of Escherichia coli to perform repair resynthesis after ultraviolet (UV) irradiation. Purified DNA damaged by up to 100 J of UV radiation per m2 was treated with an endonuclease from Micrococcus luteus that introduces single-strand breaks in irradiated DNA. This DNA was then used as ...
متن کاملEndonuclease from Micrococcus luteus which has activity toward ultraviolet-irradiated deoxyribonucleic acid: its action on transforming deoxyribonucleic acid.
An endonuclease purified from Micrococcus luteus makes single-strand breaks in ultraviolet (UV)-irradiated, native deoxyribonucleic acid (DNA). The purified endonuclease is able to reactivate UV-inactivated transforming DNA of Haemophilus influenzae, especially when the DNA is assayed on a UV-sensitive mutant of H. influenzae. After extensive endonuclease action, there is a loss of transforming...

متن کاملMutagenic action, loss of transforming activity, and inhibition of deoxyribonucleic acid template activity in vitro caused by chemical linkage of carcinogenic polycyclic hydrocarbons to deoxyribonucleic acid.
In a solvent system of 10(-2)m phosphate buffer (pH 6.8)-ethanol (2:1, v/v) and in an iodine-induced reaction, the polycyclic hydrocarbons [(3)H]3,4-benzpyrene and [(3)H]3,4-BP/[(3)H]9, 10-dimethyl-1,2-benzanthracene (DMBA) can be covalently linked to deoxyribonucleic acid (DNA) at room temperature. By stepwise addition of the hydrocarbon and repeating the reaction two to three times after isol...
